Hi, I already bought the E50 II and the PA5 II with balanced cables. Am I missing out on much with this new Mini 300?
You're not missing anything, except that Topping has managed to offer comparable performance and measurements in a less expensive device.
The Topping PA 5 II uses a few higher-quality and more expensive components, but these don't result in a longer lifespan or audibly better measurements.

I would argue that you're not missing out on anything audible.

Things to try with this:
1. Run balanced, but use - for hot and reverse the speaker cables. The reason is that many cheap opamps have poorer PNP transistors than NPN. Going in backwards (And fixing it on the outputs) uses the NPN transistors. Made a HUGE difference with my Hypex 122 amp because it's input chip is a 5532.

That won't work with the Mini 300; you'd have to modify the inputs and outputs.
The TPA3251 in the Mini 300 is already configured for BTL; for mono, you'd have to change the circuit to PBTL. It's best to take a look at the datasheet.

Even if they painstakingly convert the Mini 300 to mono operation, they won't increase the output power (it's already configured for BTL), but will only make it 2-ohm capable, even though it's not very sensitive to that anyway.

However, a 3E Audio A5 with 2 x TPA3251 in dual mono PBTL operation would be the better choice and roughly the same price.
